Effect of fluoride in bone repair of rat alveoli: MMP-2 and 9 and BMP-2 and 7 proteins analysis

Abstract

There are several factors and modulators that influence bone formation, fluoride being among them for besides it is considered an essential element due to its beneficial effects on health it also stands out by influexing the osteoblasts activity, which are responsible for intermediating bone syntheses and resorption (interacting with osteoclasts). This way, the present paper aims to evaluate alveolar bone repair in rats under the effect of fluoride provided in drinking water. In this work it will be used 4 groups of Wistar rats (Rattus norvegicus) 80 days old, who will receive deionized water and water with 5, 15 and 50 fluoride ppm. There will be made surgical procedures to extract right upper incisor and later on, the animals will be euthanasied 7, 14, 21, 30 and 60 days. Imediately after euthanasia, it will be made a heart punction to obtain blood plasma for fluoride analysis and to obtain the piece with the right tooth alveoli. Microscopic analysis will be made (colored in hematocilin Eosin and Imunohistochemical) and of proteins related to bone repair (MMP-2 and 9, and BMP-2 and 7 proteins) through Western blot technique. Those analysis can provide details of new bone formation in animals who received or not, fluoride treatment. (AU)
