Study of bone repair in dental alveoli of rats under the effect of fluoride: analysis of transcripts BMP-2 and 7 and MMP-2 and 9

Abstract

The comprehension of molecular mechanism of bone repair and the influence of some factors on the repair may help in the treatment of fractures and bone defects. Fluoride is an element in the daily life of almost any Brazilian population, being used in water supply (Brazil) and also as therapy for osteoporosis (commonly in Europe). Therefore it is important to understand the influence of fluoride in the physiological repair of bone tissue. This study will be used with 4 groups of rats Wistar of 80 days (n = 100), which receive drinking water containing 5, 15 and 50 ppm of fluoride, and deionized water (control) throughout experiment. These animals have the right upper incisor extracted. The animals will be killed 7, 14, 21, 30 and 60 days after the extraction and collect of material for analysis of proteins related to repair bone (bone morphogenetic protein - BMP-2 and 7, the matrix metalloproteinases - MMP-2 and 9, and alkaline phosphatase), by zymography and RT-PCR. After the preparation of the protein tissue reaction of rats, they will be subjected to two-dimensional electrophoresis for analysis zymography. The gels of treated and control groups will be scanned using the ImageScanner and analyzed with Kodak Molecular Imaging Software ®. Will be investigated the mRNA for MMP-2 and 9, BMP-2 and 7 and Alkaline Phosphatase by real time RT-PCR. The parametric analysis is performed using analysis of variance (Tukey test was p <0.05). (AU)
