Quantification of enterobacteria and Clostridium spp. and molecular detection of Clostridium perfringens, Escherichia coli and Salmonella spp. at points in the chicken meat production chain

Abstract

Brazilian poultry production has been increasing significantly, mainly by the consumption of chicken meat, which leads to concern about the transmission of pathogens to human. However, proper hygiene and management in the production chain of poultry meat may avoid food bacteria contamination. Thus, this study aimed to evaluate the presence of pathogenic bacteria in poultry farms, sanitary conveyors of Brazilian slaughterhouses, before and after the pre-operational and operational hygiene, and in frozen cuts of chicken commercialized in supermarkets, beyond of comparing isolates obtained in different points of the production chain by repetitive sequences using Rep-PCR. The same isolates were also submitted to antimicrobial sensitivity test. For this, it was performed Enterobacteriaceae and Clostridium spp. counts, identification of Salmonella spp., Clostridium perfringens and diarrheagenic Escherichia coli (Enteropathogenic E. coli - EPEC, Shigatoxigenic E. coli - STEC, Enteroaggregative E. coli - EAEC, Enterotoxigenic E. coli - ETEC) through PCR and bacterial isolation. For quantification of Enterobacteriaceae and Clostridium spp. and PCR identification of pathogens, 311 samples were collected from sanitary conveyors using sterile swabs. From poultry farms, 164 samples of cloacal content swabs were collected from healthy chickens, and 20 samples of frozen chicken cuts were taken from supermarkets. In commercial poultry farms, 107 positives samples for EPEC were identified. Similarly, it was found 23 positive samples for EPEC and one for C. perfringens in sanitary conveyor at chicken slaughterhouses. In cuts of chicken from supermarkets, it was found four positive samples for EPEC, two for STEC and four for C. perfringens. No samples were diagnosed with the presence of Salmonella spp.. The genetic profile analysis of 27 EPEC strains showed that there was a genetic relationship among isolates from the same origin and among those obtained from different points of the production chain, which is an indicative of cross contamination. All isolates are resistant to at least one antimicrobial. Thus, according to the results obtained, it was attested that bacterial contamination might be present in various points of the poultry meat production chain, from the poultry farms to the final product. Certainly, it denotes the importance of enhanced care with the hygiene and management in all stages of the meat production, which possible will reduce the chance of these harmful pathogens transmission to human.
