The Feasibility of the Alcohol-Based Deep Eutectic Solvents: From Thermophysical Characterization to Application in Active Pharmaceutical Ingredients Systems

Abstract

Deep eutectic solvents (DES), a green solvent class, are of interest for the chemical and pharmaceutical industry, are composed of a hydrogen bond acceptor (HBA), usually a quaternary ammonium salt, and one or more hydrogen bond donors (HBD). The physicochemical properties of the DES depend on the type and proportion of their constituents. In this work, DES composed of choline chloride, as HBA, and polyols (glycerol, ethylene glycol, or 1,2-propanediol), as HBD, were obtained at three different molar ratios. The DES' physical properties, namely: density, viscosity, and speed of sound, were measured at different temperatures (from 298.15 to 353.15 K). The temperature and HDB chain size effects on these properties were evaluated. Lastly, the proposed DES were assessed as a potential vehicle for processing active pharmaceutical ingredients (APIs). The DES properties are demonstrated to be dependent on both temperature and HBD nature. Applying to API processing, the proposed DES significantly increased the APIs solubility compared to the pure water and promoted the separation/purification of these compounds in aqueous biphasic systems (ABS). The solubility and partition assays were supported by the in silico tool COSMO-SAC model (conductor-like screening model for real solvent segment activity coefficient). Finally, the results suggest the necessity to know the solvent's physical properties of interest and the versatility and applicability of the DES on the pharmaceuticals industry. [GRAPHICS] . (AU)
