Abstract

Purpose:* To culture and to characterize lateral elbow epicondylitis cells with the markers: collagen type I, II, III, scleraxis, osteocalcin, alpha muscle actin and vimentin by immunohistochemistry.* To evaluate the basal production index of inflammatory cytokines IL-1², IL-6, IL-8, IL-10, TNF-±, FGF, VEGF and GM-CSF by lateral epicondylitis cells in cell culture.* To compare the in vitro anti-inflammatory effect on the production of inflammatory cytokines on lateral epicondylitis cells after exposure to triamcinolone and PRP.Methods:An in vitro study will be performed with primary culture of lateral epicondylitis elbow cells. The cultured cells will initially be characterized by the immunohistochemical marking with collagen type I, II and III, scleraxis, osteocalcin, alpha muscle actin and vimentin, in order to elucidate the cellular composition of the lateral epicondylitis in tenocytes, osteoblasts or fibroblasts.Subsequently, a MTT viability assay will be performed after exposure to different concentrations of triamcinolone acetate, in order to establish the concentrations and time points to be studied. Cells of third passage will be exposed in triplicates to triamcinolone and platelet-rich plasma for comparison of their anti-inflammatory effects, accessing the production of the cytokines IL-1², IL-6, IL- 8, IL-10, TNF-±, FGF, VEGF and GM-CSF by ELISA. The study of soluble SGP-130 receptors will indicate whether IL-6 is exerting an anti-or pro-inflammatory effect. The data will be submitted to statistical analysis by parametric test not matched with Welch's correction. (AU)
