Abstract

The platelet-rich plasma (PRP) is an autologous product obtained from the whole blood, which has been clinically used in various fields of regenerative medicine, because of its ability to stimulate tissue regeneration. In the modern classification of the blood concentrates, the P-PRP (pure-PRP, which is rich in platelets and poor in leukocytes) and L-PRP (rich in platelets and leukocytes) are the most commonly used in clinical therapies. Recent studies have shown that the balance between anabolic effects (platelets) and catabolic effects (leukocytes) in L-PRP is responsible for the regulation of cellular events on tissue regeneration. Hyaluronic acid (HA) is a glycosaminoglycan present in soft tissues and joint fluid. Its importance in biological process includes all molecular weight range, 103-106 Da. Clinical uses of L-PRP and HA of high molecular weight have shown many benefits in bone tissues regeneration. However, fundamental studies in vitro are still scarce and therefore, necessary to characterize their association and biological effects. In this context, the present work aims to prepare and characterize the association between L-PRP/HA as a new product, and investigate their effects on human adipose-derived mesenchymal stem cells (h-AdMSCs) proliferation and osteogenic/ chondrogenic differentiation, aiming standardization. Furthermore, this project proposes the use of microfluidics technology for cell migration studies, in response to L-PRPs releasate gradients. The work should be conducted in the LDPB/UNICAMP with the collaboration of the Khademhosseini Lab-Harvard/MIT, USA. L-PRPs of three ratios platelets/ leukocytes (>1, =1 e <1) and HAs of 103, 105 e 106 Da will be studied. This work has technological (preparation of autologous product L-PRP/ HA) and clinical (orthopedic applications) relevance. (AU)
