Abstract

Platelet Rich Plasma (PRP) is an autologous therapy, non-immunogenic, which can stimulate and accelerate tissue regeneration. Despite the intense use of this therapy in orthopedic medicine, scientific data related to analysis and /or standardization of PRP preparation are still very dispersed. The prepared PRP's quality is essential to the success of its clinical application. Critical aspects such as, the early release of its growth factors during PRP preparation, making them inactive, and the fast in vivo release of GF reduce the clinical efficacy of PRP therapy, since the bone regeneration process requires a GF release with spatial and temporal control. Seeking to improve these aspects, this research project proposes the modulation and optimization of PRP quality as a function of the operational variables involved in the overall preparation process, and its encapsulation in hydrophilic gel as strategies for PRP standardization, protection and gradual in vivo release of their GF. In the first case, the study will use the statistical factorial design and response surface analysis, applied to the PRPs preparation with and without leukocyte layer. The encapsulation will use hyaluronic acid hydrogels in the form of particles, ionically crosslinked with calcium gluconate or polylysine. Both are unpublished. The results of this project will contribute to PRPs safe and reproductive preparation, and to increase their effectiveness in clinical applications. (AU)
