Abstract

The endocrine and nervous systems interact reciprocally with peripheral organ and tissues in the regulation of several body functions. Homeostatic responses are activated by peripheral signals, such as metabolic factors, glucose, FFA, electrolytes, hormones, cytokines, and myokines that interact with neuronal and non-neuronal circuitries for the maintenance of internal milieu. Besides its structural function, the role of bone as a target and modulator of energy homeostasis has been well recognized. The influence of prenatal and neonatal environment as risk factors for metabolic disease in juvenile and adult life has been conceived. To investigate these aspects, the present project includes the subproject 1 hypothalamic neuromodulators involved in the hydroelectrolytic balance in response to osmolality and volume changes; subproject 2 reciprocal interaction between nervous system and mesenchyme tissues on energy and osteomineral homeostasis and subproject 3 clinical investigation of the influence of metabolic and mineral disorders on the interplay of bone, muscle and adipose mesenchymal tissues. The proposal comprises experimental, translational and clinical models that will use diverse approaches, including conditional deletion (cre-lox methodology), nutritional deprivation, overnutrition (changes in litter size), analysis of body composition, transcriptome analysis of hypothalamic tissue, chemogenetic (DREADD), optogenetics, gene silencing (lentiviral and AAV microinjection), cell morphology study and cell culture. Therefore, the proposal will provide relevant original data and contribute for a better understanding of central and peripheral mechanisms involved in the complex interaction of mesenchymal tissues on energy and osteomineral homeostasis.
