Analysis of the neuroprotective activity of some chlorogenic acids present in the extracts of plants from the genus Lychnophora (Asteraceae; Asterales)

Abstract

Chlorogenic acids, commonly found in asteracean species, have been shown to exert anti-inflammatory effects in a wide range of in vitro and in vivo experimental models. These compounds can be considered as candidates to neuroprotective agents due to the strong correlation between oxidative stress and neuronal damage. In this context, the use of anti-oxidant and anti-inflammatory agents may represent an alternative co-adjutant therapy to prevent neuronal tissue damage which occurs as a consequence of ischemic processes, Status Epilepticus (SE) and brain trauma. In the light of these facts, the present work is aimed at the investigation of the neuroprotective effects of some chlorogenic acids isolated from extracts of L. ericoiddes against neuronal damage induced in Wistar rats submitted two different experimental models. In the first group, animals will receive i.c.v. injections of pilocarpine in order to induce a SE and consequent death of neurons in the limbic system. In this experiment, electroencephalographic and behavioral alterations will be monitored by video EEG before, during the SE (3h), after attenuation by thiopental injection and after the administration of chlorogenic acids. Animals in the second group will receive injection canulae in the anterior chamber of the eye in order to increase intra-ocular pressure. In this experiment, different concentrations of chlorogenic acids will be delivered directly into the aqueous humor. In both experiments, histological alterations will be analyzed in a quantitative and qualitative manner by histological routine techniques. Moreover, the expression of neuronal lesion and proliferation markers will be monitored by immunohistochemistry and imunofluorescence. (AU)
