Abstract

Mineral trioxide aggregate (MTA) has been used in various endodontic applications. Despite the advantages of MTA, some drawbacks may limit its use, including high cost, physical and chemical properties, and difficulty in handling. In this context, a novel calcium aluminate cement, composed of Al2O3, CaO and additives, has been developed to overcome some negative MTA characteristics. The aim of the present study is to evaluate different parameters of the progression of primary osteogenic cell and of dental pulp-derived stem cell (OD-21 lineage) cultures exposed to different preparations of calcium aluminate cement using MTA as a control. The following parameters will be assessed: 1) dissolution of the element calcium in the culture medium at 3 days, 2) cell morphology at 1 and 3 days, 3) cell viability, proliferation index and total cell number at 1, 3, 7, and 10 days, 4) immunolocalization of extracellular matrix proteins at 1, 3, and 7 days, 5) mRNA expression for genes related to osteoblastic and odontoblastic differentiation at 7 and 10 days, 6) alkaline phosphatase activity at 3, 7 and 10 days, 7) proportion of cells undergoing apoptosis at 10 days, and 8) matrix mineralization at 14 days.
