Effect of non-digestible oligosaccharides FOS and GOS on the intestinal and bone metabolism of female Wistar rats on different years of age

Abstract

Prebiotics are non-digestible oligosaccharides (NDOs) fermented by intestinal bacteria that can induce health benefit effects on the host associated. These benefits effects can be local or systemic, as for example, changes on intestinal flora, improving minerals absorption and bone metabolism. Young mammalian animals start to compound their intestinal flora after were born and improve their bone calcium content in the early stage of life too. Further, there are many physiologic mechanisms aging related that can change intestinal flora and improve bone loss. Thus, the objective of this study is to evaluate the effect of NDOs FOS and GOS on the intestinal and bone metabolism of female Wistar rats with different years of age. Thirty-two female rats with 21 and 270 days of life will be fed with AIN-93G and AIN-93M, respectively, containing FOS, GOS and FOS/GOS per 60 days (Group 1 = standard diet; Group 2 = FOS diet; Group 3 = GOS diet; Group 4 = FOS and GOS diet). The effect on intestine will be evaluated by fecal pH determination, SCFA and Bifidobacterium e Lactobacillus content, cecal histomorphometry and gene expression; bone metabolism will be evaluated by dual-energy X-ray absorptiometry, histomorphometry, biomechanic properties and serum biochemical assays. All data will be analyzed by GraphPad software 5.0 with two-way ANOVA and Tukey's test (p<0,05). (AU)
