Gene expression in individuals with type 2 Diabetes mellitus , dyslipidemia and periodontal disease: evaluation by microarray

Abstract

The present study aims to investigate gene expression in patients with type 2 diabetes (DM2), dyslipidemia and periodontal disease by microarray technique. The development of this project is in order to complement another study that is already being conducted at Araraquara School of Dentistry (FOAr/UNESP), under the responsibility of Profa. Dra. Silvana R. Perez Orrico, who intends to evaluate the influence of lipid peroxidation on inflammatory markers in patients with type 2 diabetes and chronic periodontitis. The subjects that will be investigated in the present study are the same individuals selected in the above mentioned study and have been subdivided into 3 groups: Group 01: patients with dyslipidemia, periodontal disease and diabetes with adequate metabolic control, group 02: patients with dyslipidemia, periodontal disease and diabetes with inadequate metabolic control and group 03: patients with dyslipidemia, periodontal disease and without diabetes. It is also intended to include in the present study two additional groups, being group 4: with periodontal disease, without diabetes and dislipidemia; and group 5: without diabetes, dyslipidemia and periodontal disease. The groups 1 and 2 with diabetes, have being evaluated by the percentage of glycated hemoglobin (HbA1c), being considered with inadequate metabolic control when HbA1c is > 8.0%. All the patients have been through complete periodontal disease, physical evaluation and laboratorial evaluation of plasma fasting glucose, lipid profile and levels of lipid peroxidation (oxidized LDL and malondialdehyde - MDA). Blood collected from each patient have been submitted to leukocytes separation to investigate gene expression. From each group, 5 patients will have the gene expression evaluated by microarray technique, whose results will be analyzed by bioinformatics and biostatistics. Then, it will be selected 3-4 genes differently expressed that may play a role in the pathophysiology of the investigated alterations. It will be validated by Real time PCR, using the TaqMan system, using two genes of constitutive expression (endogen controls) to normalize the gene expression data.It is important to emphasize that the application of the microarray methodology in the present study will contribute to a global vision of the gene expression pattern, influenced by diabetes with an inadequate metabolic control and by the presence of dyslipidemia and/or periodontal disease. Besides, this study will propitiate an important partnership between researchers from different departments of the Araraquara School of Dentistry, and from different Universities such as the Medicine School of Ribeirao Preto - São Paulo University (USP) and State University of Campinas (UNICAMP).
