Isolation, molecular identification and toxigenic potential of fungi and occurrence of mycotoxins in cereal mixtures traded in Brazil

Abstract

The aim of the project is to isolate and identify fungi, evaluate the toxigenic potential of Aspergillus spp. and Fusarium spp. isolates, detect and quantify aflatoxins (AF) B1, B2, G1, G2 and fumonisins (F) B1 e B2 in samples of cereal mixtures denominated "ração humana". It will be analyzed cereal mixtures ready to eat, acquired from companies which trade the products nationally by internet. Samples of 15 different marks will be evaluated monthly, for a period of seven months, adding up 105 samples in the end of experiment. The unit sample will be constituted for original package with minimum of 250 g. Fungi from Aspergillus genus will be classified to specie level by genetic sequencing. Mycotoxins determination will be performed by high performance liquid chromatography, using immunoaffinity columns to clean up samples, following the evaluation of the method by performing analyses in samples artificially contaminated with toxins. Results will allow the knowledge of the contamination of the cereals mixture by toxigenic fungi and mycotoxins, and the simultaneous occurrence of aflatoxins and fumonisins in the products. (AU)
