Interactions between different Candida species in experimental candidiasis of invertebrates and vertebrates models

Abstract

Whereas the high incidence of lesions of candidosis in immunocompromised patients and the emergence of non-albicans Candida species, which are often isolated in association with C. albicans in mixed infections, it becomes necessary to develop in vivo models in order to characterize the interactions among Candida species. This study will evaluate in vivo interactions between different Candida species by inducing experimental candidiasis in Galleria mellonella and immunosuppressed mice models. Standard strains of C. albicans (ATCC 18804), C. tropicalis (ATCC 13803) and C. glabrata (ATCC 90030)will be used. The larvae of G. mellonella will be inoculated with heterotypic and homotypic suspensions of Candida (105 cells / mL) and incubated for 5 days. The number of dead larvae will be evaluated daily and the survival curve and statistical analysis will be performed in the program Graph Pad Prism using the Log-rank test (Mantel-Cox). For the induction of oral candidiasis in mice, the animals will be treated with tetracycline hydrochloride (0.83 mg/mL) in drinking water and injections of prednisolone at a dose of 100 mg/kg body mass. The animals will be inoculated with 0.2 ml of homotypic or heterotypic microbial suspensions containing 108 cells/mL. After 24 hours the inoculation, samples of the tongue will be collected and seeded in HiCrome chromogenic agar for counting colony forming units per ml recovered from the oral cavity. Then the animals will be euthanized and their tongues removed for macroscopic and microscopic analysis. Data analysis of CFU/mL will be made by ANOVA and Tukey test. For the evaluation scores of macroscopic and histological analysis will be applied the Kruskal-Wallis and Bonferroni tests. (AU)
