Abstract

The most effective pain control has motivated the research for new local anesthetic formulations. The association with lipossomal or ciclodextrin carriers has been proposed in order to prolong the anesthetic effect and reducing its toxicity. New anesthetic formulations produced a low systemic toxicity and longer duration of anesthesia. Adverse effects were also observed. One of the side effects was the appearance of inflammatory painless process without apparent cause. This inflammatory response of unknown nature could be triggered by the release of proinflammatory mediators. This study aims to check the effects of local anesthetics and associated with carriers on the modulation of inflammatory mediators IL-6, IL1-±, IL-8, TNF-±, IL-10 and PGE2, in addition to its action on cell viability and Caspase-3 production in cultured oral epithelial cells and human gingival fibroblasts. The cells will be exposed to different concentrations of anesthetics bupivacaine, lidocaine, ropivacaine, and benzocaine free or associated with liposomes or cyclodextrin. Quantification of cytokines and PGE2 will be done using the capture ELISA after 6h and 24h. Cell viability will be estimated after the exposure period and compared to the untreated control group. Statistical analysis will be performed by ANOVA (Tukey test) or Kruskal-Wallis test (Dunn), BioEstat 5.0 statistical package (AU)
