Abstract

Adipose tissue macrophage infiltration and pro-inflammatory adipokine production contribute to chronic low grade inflammation observed during obesity and it initiates peripheral tissue insulin resistance. The increase in free fatty acids level due insulin resistance in adipose tissue seems to be responsible to the accumulation of free fatty acids in muscle, liver, pancreas and heart resulting in pathologies like type 2 diabetes. Adenosine levels are increased during inflammation and it can modify metabolic and inflammatory response. Adenosine receptors A1 and A2A mediate pro- and anti-inflammatory response, respectively. A2A activation results in TNF-a inhibition, an important mediator produced by adipose tissue that was a role in the establishment of insulin resistance. A1 receptor activation also mediates an anti-lipolytic response and the lipolysis inhibition controls free fatty acid release and metabolism during obesity. Adenosine also modifies adipogenesis and tissue remodeling, probably controlling adipose tissue expansion. The aim of this project is to study the adenosine receptors activation role in adipose tissue alterations during obesity focusing metabolic, morphological and inflammatory alterations. We intend use selective and unselective agonists for adenosine receptors as well as high-fat diet induced obesity model in mice and 3T3-L1/RAW macrophage cultures as in vitro obesity models. (AU)
