Abstract

In the pharmaceutical industry, new medicinal products using nanotechnology are proposed in order to increase the therapeutic index, decrease adverse effects, aim drugs to specific sites, reduce toxicity and controlled release of the drug. Researches in this area are aimed at improving the treatment of diseases with high incidence and complexity, such as cancer and parasitic infections, viral diseases, among others. This project proposes the use of techniques antisolvent supercritical gas (SSA) and the rapid expansion of supercritical solution (ERSS) for obtaining and evaluating of gastroretensives and floating solids systems and solid dispersions. The particles will be obtained through both techniques and will be carried out in different supercritical conditions using the same equipment. The products from the SSA and ERSS techniques will be evaluated for variation in size and particle morphology, chemical interaction between components of the formulation, physicochemical properties of solid, level and profile of drug release. These characteristics will be investigated by dispersive X-ray low angle (SAXS), scanning electron microscopy (SEM), infrared spectroscopy with Fourier transform (FTIR), thermal analysis (DSC), X-ray diffraction (DR-X) and the dissolution study. The results of this study will be an advance in the generation of nano and microstrutured therapeutic systems and to increase bioavailability of drugs, whose bioavailability is limited by the rate of dissolution. Besides being a cleaner technology that conventional method, the others advantages of supercritical fluid technology are the moderate conditions of the process, elimination of residual solvents and easy scale-up.
