Abstract

This project aims at updating and extending the Thematic project developed during the past 5 years (F APESP No. 95/9344-4). The General Objective is to reveal the molecular bases for normal/regulated processes of cell proliferation and differentiation and for the escape from this control that leads to tumor growth, using in vitro and in vivo transgenic systems (cell cultures and animals) in addition to cutting edge technologies of Genomics, Proteomics, Tissue Engineering and Bioinformatics. The following Specific Objectives are proposed: 1. expression analysis of genes associated with control of cell proliferation and differentiation and the origin of neoplasia; 2. searching, isolation and structural characterization of genes which are differentially expressed in the presence of mitogenic agents, differentiation-inducers, tumor virus and anti-tumor/anti-proliferative agents; 3. in vitro functional analysis of genes isolated in the previous item; 4. in vivo functional analysis of genes isolated in item 2, generating "knock-in" and "knock-out" transgenic animals; 5. isolation, culture, characterization and genetic modification of cells obtained from human and animal tissues and organs; 6. analysis and isolation of new genes from human and animal origin; 7. production of recombinant proteins of scientific and biotechnological interest; 8. generation of specific polyclonal antibodies to proteins of interest. The viability of this project is based on a consolidated Team of about 30 people, distributed in 2 well equipped laboratories (Cell and Molecular Biology Lab. and the Human Pancreatic lslet Unit) and on extensive networking with other Brazilian and foreign research groups. ln addition to generating new and relevant knowledge, we hope to continue to yield well-trained people who are qualified to use the knowledge generated in the project to develop technologies and to formulate new strategies for Cell and Molecular therapy of diseases.
