Reproductive aging in dogs: clinical and biotechnological features

Abstract

Recently, aging is considered a research goal in several small animal specialities. Increase in canine longevity is verified over the years, similarly to men. On the other hand, few studies have been performed, imparing medical treatment of the senile dog. The present research will be divided into separated experiments: Experiment 1 - differences in fresh and cryopreserved sperm profile of young and senile dogs; Experiment 2 - Phisiopathological changes of the hyperplasic prostate of dogs subjected to orchietomy or finasteride treatment; Experiment 3 - influence of finasteride on the prostate and testicles of dogs with Benign Hyperplasia Prostate (BHP). In experiment 1 semen will be collected from young and old dogs, composing two experimental groups. Each group will be additionaly subdivided into Fresh and Cryopreserved semen. Dogs of the experiment 2 will be diagnosed with BHP and will be randomly divided into the following groups: Orchiectomy (n=5), Orchiectomy+Finasteride (n=5), Finasteride (n=5) and Control Group (n=5). In experiment 3, dogs will be divided into 4 groups: Control (n=5), Control Finasteride (n=5), BHP finasteride (n=5) and BHP Group (n=5). The sperm rich fraction will be collected and submitted to imediate analysis of motility, vigor and sperm count, functional sperm tests (plasma membrane and acrosomal integrity, mitochondrial activity and DNA fragmentation), antioxidant profile, protein oxidation analysis, AGE and carnosine concentrations. Dogs of experiment 2 and 3 will be analysed by doppler ultrasonography of the prostate and testis, testosterone, estrogen and di-hydrotestosterone assay, imunoistoquimical and real time PCR analysis of VEGF of prostate parenchyma, obtained by biopsy.
