Abstract

Venomous animals are amongst the most successful animals on Earth's evolutionary history. One of the important factors for their success is the production of venoms, used to subdue prey and as protection against possible predators. Most active toxins are proteins and peptides, elaborated and refined over millions of years of evolution. Venom toxins have served as leads for molecules with potential pharmacological and biotechnological applications. In addition, they have been important in elucidating biochemical and physiological mechanisms in vertebrates. A landmark in venom research was the discovery of the bradykinin-potentiating peptides, which eventually led to the development of the first drugs for the treatment of arterial hypertension. The development of analytical tools such as next-generation sequencing and mass spectrometers have resulted in the elucidation of thousands of new toxin sequences in recent years. However, despite the large number of sequences discovered, and the maturity achieved by proteomic studies, venom peptidomics is still an emerging research field. Particularities such as the lack of enzymatic specificity in the peptide search and the absence of databases create analytical challenges for such studies. Our group has been working on the improvement of mass spectrometry based analytical techniques applied to the peptidomics, which resulted in the elucidation of several new biologically active venom peptides. We also observed that small mutations have significant effects on the biological activities of peptides within the same class. Given the richness of the still largely unexplored peptidomes venoms, we propose in this project the peptidomic analysis of Brazilian snake and spider venoms with the objectives of unraveling novel peptide sequences, map mutations between homologous sequences and prospect potential biological activities of these peptides.
