Abstract

The study aims to validate the methodology of stable carbon isotopes to determine the gastrointestinal transit time in pacu (Piaractus mesopotamicus), in contrast to the addition of contrast markers in the diet in order to find a more accurate and reliable for this analysis. The test will be conducted at the Aquaculture Center of UNESP (CAUNESP), Campus de Jaboticabal. Will be used 288 fingerlings weighing 50 ± 10 g distributed in 36 aquariums with a capacity of 150 L. The experiment will be carried out with eight treatments and four replications in a 2x2x2 factorial corresponding to two growth temperatures (25 °C and 29 °C), two diets (one based on C3 plants and other based plant C4) and two methodologies for determining the gastrointestinal transit time (use of inert markers and stable isotopes). The fish are fed twice a day for a period of 90 days. To determine the gastrointestinal transit time will be carried faeces samples over 24 hours at intervals of 30 minutes. For the treatment of the methodology that uses contrasts of inert markers (1% titanium oxide and then 1% of chromic oxide), the faeces will scores from 0, 0.25, 0.50, 0.75 and 1.00 according to the intensity of change staining. For the treatment of stable isotope methodology, the faeces will be analyzed to determine the change in the isotopic ratio of carbon-13 the trial period to evaluate the gastrointestinal transit time. At the end of the analysis, the results between the two methods are compared to show the accuracy and possible errors associated with traditional methods of using markers of contrast in the diet as well as the possible interference of different temperatures. The data will be submitted to analysis of variance (ANOVA) and when significant differences, the averages are compared by Tukey test at 5% probability.
