Abstract

Glioblastoma (GBM),IV astrocytoma (WHO), primary brain tumor is the most frequent malignant in adults. A tumor characterized by being highly resistant to therapy, and since the poor prognosis of patients with glioblastomas and current treatment, new therapeutic approaches are needed. One attractive approach is the use of specific tumor antigens as targets to stimulate the patient's immune system. A prerequisite for the success of these strategies is the existence of genes that are exclusively or preferentially expressed in malignant tissues compared with normal tissues. Cancer-testis antigens (CTA) are considered as potential candidates for development of vaccine, because they are expressed in various tumors and their expression in human tissues is nearly restricted to germ cells of the testis and trophoblast cells. Allied to the fact that many of the findings present in placentation and gametogenesis, such as migration, invasion, immune escape, resistance to apoptosis and induction of angiogenesis are the same ones that give their malignant potential tumors, it is very important to understand the cellular function of these CTAs in tumor cells. So from the functional analysis of four genes validated CTAs with high expression in glioblastomas, not expressed in normal brain tissue is intended to: examine different cultures of GBM cell lines the overexpression and silencing the four CTAs; check for correlation with the expression of these antigens in GBM cell lines and proliferation, apoptosis, invasion, migration and cell cycle changes.
