Abstract

The most frequent cause of recessive hearing loss are mutations in GJB2 gene (connexin26, Cx26). Among the individuals with hearing loss with mutations in this gene, 10% to 50% present a single recessive mutation in heterozygosis, indicating that haplo-insufficiency of GJB2 may interact with other types of mutations in the same gene, in the neighbor gene GJB6, or even in other genes. The auditory system is complex and specialized. Physical and functional interactions among different proteins are necessary to the physiology of hearing. Most studies with Cx26 are related to the gene encoding the protein and its pathogenic variants, being few proteins known to interact physically and functionally with Cx26. During the doctor program, we searched for Cx26 interacting proteins by pull-down assays. We Identified a total of 22 candidate proteins for interaction with the C-terminal region of Cx26. Since confirmation of the interaction between these 22 proteins and Cx26 is desirable through studies of immuno-coprecipitation, followed by studies of cellular and subcellular localization by immunofluorescence and functional studies, these will be our goals in this postdoctoral proposal.
