Effects of training and overtraining on the content and activation of proteins of insulin signaling pathway in the skeletal muscle of Swiss mice

Abstract

Based on the relationship between overtraining and high muscle contents of tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6), and knowing that these cytokines are associated with the impairment of the insulin signaling pathway, the main aim of the present research project was to verify the effects of overtraining on the content and activation of the proteins of an insulin signaling pathway (IR-beta, IRS 1, IRS-2, PI3q, and Akt), on the pro-inflammatory proteins SAPK/JNK and IKK-beta, and on the content of SOCS3 protein in skeletal muscle of Swiss mice. The mice will be shared on three experimental groups: Control (C; sedentary), Trained (TR; mice will be submitted to the training protocol), Overtraining (OT; mice will be submitted to the overtraining protocol). The body weight and food intake of all groups will be recorded weekly. The mice will perform the insulin tolerance test at the end of the seventh week. At the end of the eighth week, 24 hours after the exhaustive test, mice from C, TR, and OT will be sacrificed. The content and activation of the proteins mentioned above will be measured on the extensor digitorum longus and soleus by immunoblotting. Results will be expressed as mean ± standard error (SE). The analysis of variance will be used to examine the effects of training and overtraining protocols on the studied parameters. When analysis of variance indicates significance, Bonferroni's post hoc test will be performed. The significance level will be 5%.(AU)
