Abstract

The aim of this study is to evaluate the relation between diabetes mellitus, tissue response and the mineralization ability of sealers Gray MTA Angelus®, White MTA Angelus®. Zinc Oxide and Eugenol® will be used as a reference of less biocompatible cement. Twenty-four rats will be divided into two groups of twelve animals each: group 1: normal rats, group 2: diabetics rats induced by Alloxan. All animals receive four implants of polyethylene tubes, three containing sealers to be tested and an empty tube for control. At 7, 14 and 21 days will be carried out injections of fluorochromes (Calcein, Alizarin and Oxytretacycline) .After 7 and 30 days, six animals of each group will be sacrificed and the tubes with surrounding tissues removed and fixed. The tubes will be cut in half and a portion processed and included in glycol methacrylate, which are observed in HE staining, Von Kossa, without staining for analysis with polarized light and fluorescence. Tissue reactions on the opening of the tube in contact with the material will receive scores as 0, none or few inflammatory cells and no reaction; 1, less than 25 cells and mild reaction; 2, between 25 and 125 cells and moderate reaction; and 3, 125 and more cells and severe reaction. Fibrous capsule will be considered thin when the thickness be <150 µm and thick at >150 µm. Calcification will be recorded as present or absent. The other half of the tubes will be placed in paraffin for immunohistochemical analysis for the presence of Osteocalcin, Osteopontin and Runx-2. The evaluation criteria will immunostaining: high (near by 3/4 of the cells immunoreactive (IR) per microscopic field), moderate (near by 1/2 IR cells), low (near by 1/4 cell IR) and null (absence IR cells). The results will be analyzed by ANOVA and Kruskal Wallis tests (± = 0.05). (AU)
