Effects of implantation of biosilicate scaffolds on consolidation of bone critical-sized defects

Abstract

Fractures are responsible for high rates of mortality and morbidity among affected individuals. The consolidation process in these individuals becomes impaired, which may result in delayed regeneration or even in non-union.In this context, several studies suggest that the bioactive materials are able to stimulate osteoblast proliferation and osteogenesis at the fracture site, accelerating the consolidation process. However, there is still a great need for investigation of materials which could serve as effective temporary substitutes tissue in critical-size bone defects. In this context, this project aims to investigate the effects of implantation of scaffolds of a bioactive material (Biosilicato ®) on bone repair process in rat calvaria. This study will use 20 rats, which will undergo surgery for bone defects circular with a diameter of 8 mm in the central region of the skull. The animals will be distributed among the groups in bone defect (GC) and bone defect with implantation of scaffolds Biosilicato ® (GB). Groups will be composed of 10 animals each and euthanasia will happen on the 15th day after surgery. To assess the effects of implanting the Biosilicato ® scaffolds in bone, will be made histological, morphometric, and immunohistochemical analyzes of the factors COX-2 and RUNX-2. Data will be analyzed using descriptive techniques and statistically by Student's t test, with significance level of 5% (p d 0.05).
