Abstract The use of a calcium prerinse before a fluoride rins has been shown to significantly increase fluoride concentration in saliva and dental biofilm. However, the anticaries effect of this combination is yet to be confirmed experimentally. Therefore, the iam of this study is to test the potential of a calcium prerinse, followed by a fluoride rinse, to inhibit enamel demineralization and enhance enamel remineralization, using an in situ model presenting dose-response to fluoride solutions. In a double blind, crossover study conducted in 4 phases of 14 days, 18 volunteers will use palatal appliances containing bovine enamel blocks (4 sound and 4 with artificial caries lesions). A 20% sucrose solution will be dropped onto the blocks either 8 (sound) or 3 (carious) times per day. Treatments will be: placebo prerinse and rinse; placebo prerinse followed by a 250 ppm F rinse; placebo prerinse followed by a 1000 ppm F rinse; and experimental group using a calcium prerinse (150 mM Ca), followed by a 250 ppm F rinse. After 14 days, biofilm formed and the enamel blocks will be collected for the analysis of mineral ion concentrations in the fluid and solids, and change in mineral content, respectively.
