Abstract

Cariniana estrellensis is one of the largest and most long-lived trees in the Brazilian Atlantic Rainforest. This species occurs in low population density (< 1 tree / ha), which makes it even more disturbing the current process of fragmentation of their habitat. The process of forest fragmentation and consequent reduction in the size of populations can immediately affect the reproductive system by increasing correlated mating, resulting in greater kinship in offspring. Due to these factors, C. estrellensis is endangered and studies of pollen flow and seed reproduction system and spatial genetic structure are fundamental to understand the effects of fragmentation on natural populations, generating information for the design of correct strategies in situ conservation and ex situ of their populations. Molecular markers are essential tools in the study of contemporary processes of seed and pollen flow in forest fragments. In this context, this research aims to investigate the rate of genetic isolation or immigration rate of pollen and seeds, the distance and patterns of pollen and seed dispersal, intra - population spatial genetic structure, genetic diversity, inbreeding levels and effective size in different ontogenetic stages (adults, seeds and seedlings) and inbreeding depression in a small, fragmented population of C. estrellensis located in Ibicatu Ecological Station, State of Sao Paulo. Adult trees will be measured (diameter at breast height), geo-referenced with the aid of a GPS and have sampled tissues (Exchange or leaves). Regenerating individuals will also be measured (height), georeferenced and have sampled leaf tissue. This latter sample is used to study the dispersion made pollen and seeds. In addition, open-pollinated seeds will be collected from 15 of the 26 existing trees on the fragment in two consecutive reproductive events. Sampling of seed will be hierarchical within and among fruits. 40 seeds of each tree will be genotyped, these being obtained from 8 fruits (5 seeds per fruit). So in total will be genotyped 600 seeds per reproductive event, totaling 1,200 seeds. Logically be listed many more fruits and seeds per tree. A seed sample is used to study the effective dispersal of pollen analysis of inbreeding depression and playback system. Inbreeding depression will be investigated during seed germination and in the field, such as seeds will be used to deploy a small progeny trial. Adult trees, saplings and seeds will be analyzed eight microsatellite loci. The study will generate valuable for the management of natural fragmented populations of the species information, contributing in breeding and genetic conservation for environmental restoration programs.
