Abstract

Chagas disease is a neglected illness with higher incidence in Latin America. Gene expression in T. cruzi is regulated mainly by post- transcriptional level, which emphasizes the importance of modifications in cromatin-associated proteins to control this process in trypanosomatids. The involvement of NO in the transcriptional regulation is well established in animal and plant cells and it is achieved by NO-dependent post-translational modifications of transcription factors or of secondary proteins related or not to epigenetic events. NO-dependent modifications of proteins from T. cruzi (nitration and S-nitrosylation) were recently described. Interestingly, an increment of nitration on tyrosine residues of histones (H2A and H4) was observed during the parasite adhesion to extracellular matrix, an important step in the parasite invasion, raising the hypothesis that NO may be involved in the control of gene expression in T. cruzi. In order to test this hypothesis, the ChIP-seq technique will be employed. ChIP-seq technique is an innovative methodology that uses next-generation sequencing to identify chromatin sequences associated with proteins of interest, generating maps of protein-DNA interactions. This project aims to identify the contribution of NO-dependent modifications of proteins related of the control of gene expression in T. cruzi using the ChIP-Seq technique. (AU)
