Abstract

As subproject of the thematic project Biota entitled "Phytotherapics standardized for the treatment of chronic diseases" this work aims to characterize the mechanisms involved in the antiulcerogenic action of the hydroalcoholic extract from leaves of Eugenia punicifolia (Kunth) DC (HEEP). From its popular indication for the treatment of wounds and inflammation, and based on previous results about their gastroprotective and anti-inflammatory effects in experiments in vivo, the aim of this subproject is to characterize the mechanisms involved in the healing action of HEEP to combat ulcerogenic factors such as: absolute ethanol, acetic acid, non-steroidal anti-inflammatory drugs and injuries derived from the process of ischemia-reperfusion. It is intended to characterize the main mechanisms involved in the curative activity of HEEP. For this, we will develop experimental models in which will be initially implanted gastric or duodenal lesions in animals and thereafter will be treated with EHEP for periods that can vary from 1 to 14 days of treatment. We will analyze, in different experimental groups, macroscopic lesion area, biochemical parameters such as antioxidant (superoxide dismutase, reduced glutathione, catalase, myeloperoxidase, thioredoxin, lipid peroxidation), anti-inflammatories (TNF-±lfa, IL-1beta, IL-6, IFN-gama and IL-10) and protein and gene expression of factors involved in the wound healing process (vascular endothelial growth factor and epithelial, cyclooxygenase 1 and 2, metalloproteinases 2 and 9, pro-apoptotic markers - caspase-3, caspase-8, caspase-9 and anti-apoptotic, Bcl-2). In the experiments described above, will be obtained parameters for analysis of possible toxic effects of HEEP through the evolution of body weight, lethality (death) and analysis of macroscopic organs and biochemical of plasma. From the evaluation of pharmacological effects of HEEP in healing of peptic ulcers, it is intended to offer scientific subsidies to the future incorporation of this species as a new option therapy of phytotherapic for the treatment of peptic ulcer disease.
