Abstract

The phenolic compounds derived from plants are widely used for therapeutic application. The ones that possess antioxidant activity are highlighted for reducing damage related to aging and the genesis of diseases. In relation to Eugenia punicifolia (Myrtaceae), it is known its anti-inflammatory action, the effect on the nicotinic-cholinergic neurotransmission and the use for the treatment of diabetes. For a better extraction and to potentiate the compounds related to these biological activity it is common to use statistical design of solvent mixtures. For this reason, in this project, it will be done the optimization of a mixture of solvents methanol, ethanol and water to obtain leaf extracts of E. punicifolia rich in phenolic and/or flavonoid compounds with a higher antioxidant and antiproliferative activity possible. The planning of the solvent mixtures will be made according to the simplex-centroid design. The extracts will be analyzed for total phenols with Folin-Ciocalteau reagent, total flavonoids with aluminum chloride and antioxidant activity by the method of radical kidnapping DPPH (2,2-diphenyl-1-picrylhydrazyl). Toxicity assays and cell proliferation will be carried out by using a colorimetric assay in the presence of MTT 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide against three cell types: MCF-7 (human breast cancer), HL60 (human promyelocytic line ) and human peripheral blood monomuclear leukocytes.
