Abstract

Macrophages are cell types involved in the progression of pulmonary emphysema and can take on different functions, depending on the stimuli they receive and the microenvironment. In the presence of interferon-³ (IFN-³) is the activation of macrophages M1, recognized as a pro-inflammatory. However in the presence of interleukin-4 (IL-4) and -13 (IL-13) is the alternative activation of macrophages M2 type recognized by exert anti-inflammatory action and good phagocytic action to microorganisms and apoptotic cells (eferocitose). Considerando the progression of emphysema is associated with a reduction of the activity eferocitose and pulmonary response in the presence of pollutants depends on the effective actions of macrophages to eliminate such particles, it is important to check the involvement of the different phenotypes of macrophages involved in emphysema development process in mice undergoing pollutants. For mice receive both the intranasal instillation of elastase induced emphysema, and for the controls groups receive saline and are exposed to polluted air, or filtered according to the separation of groups. We assess respiratory function, hemodynamic parameters, inflammatory profile in BAL, evaluating the mean linear intercept and proportions of elastic and collagen fibers in the parenchyma. We will evaluate the expression of cytokines IL-12, IFN-³, IL-4, IL-10 and IL-13 by ELISA in lung homogenate. (AU)
