Evaluation of antimicrobial activity of metabolites produced by Streptococcus mutans on Candida albicans: in vitro study

Abstract

Streptococcus mutans, often recognized as the main cause of tooth decay in the world, can be found along with Candida albicans in the biofilm. Products of secondary metabolism of the human microbiome are being increasingly studied, because of their direct impact on the host and other microorganisms as well as for their immunomodulatory functions. The aim of this study will be to extract the compounds present in the supernatant of S. mutans culture and see its effects on C. albicans, in vitro. The supernatant of S. mutans culture will be submitted to extraction with ethyl acetate and then the organic solvent will be removed in vacuum, under controlled pressure and temperature. Then, the extract will be dried and weighed, yielding the crude extract. The antimicrobial activity of the crude extract will be tested on C. albicans for in vitro biofilm formation and filamentation. Biofilms will be formed in bottom of 96-well plates for 48 hours and quantified by counting of colony forming units (CFU/mL). The C. albicans filamentation assays will be performed in 24-well plates and analyzed by optical microscopy. The results will be submitted to an exploratory analysis for selection of the most appropriate statistical test for each experiment of this study. (AU)
