Abstract

The topical application of nitric oxide-releasing biomaterials has shown to be effective for promoting the increase of dermal vasodilation and wound healing. The main aims of this project are the development of a new platform for topical NO release, based on polyethylene terephthalate (PET) meshes impregnated with polynitrosated polyester (PNPE) and the correlation of the kinetics of NO release in vitro with the results of their dermal vasodilation in vivo. The polyester meshes will be obtained from Band-aid® bandages. The PNPE will be synthesized through the polycondensation reaction between a diol and a sulfhydrylated diacid, generating a polysulfhydrylated polyester (PSPE). The PET meshes will be impregnated with a PSPE solution in acetone, dried and nitrosated via nitrous acid (HONO) and gas (N2O3). The kinetics of NO release will be characterized in real time by chemiluminescence and the topical vasodilation action of the meshes will be evaluated by laser Doppler flowmetry in healthy volunteers. The PET/PSPE and PET/PNPE meshes will be characterized regarding their morphologies by electron scanning microscopy, and thermal properties by differential scanning calorimetry.
