Abstract

beta-glucosidase are enzymes that hydrolyze terminal beta-(1.4) glycoside bonds, essential function to many biological processes, being of great interest to biochemistry and biotechnology. This study involves the extraction and subsequent purification of ²-glucosidase of thermophilic fungus Thermoascus aurantiacus in a supermacroporous support of polyacrylamide produced in cryogenic conditions and chemically modified by the introduction of ionic groups N, N-dimethylaminoethyl methacrylamide. For this, different pH values and different flow rates of the mobile phase will be tested on the specific activity of the purified enzyme. After the purification step, an immobilization procedure will be studied using the same support. The effects of pH and temperature on the stability of free and immobilized enzymes will be determined, as well as the determination of the kinetic parameters of Michaelis-Menten model. Finally, the performance of the system containing the immobilized enzymes will be tested in the hydrolysis of isoflavone and degradation of pretreated sugar cane bagasse. (AU)
