Therapeutic and preventive effect of gamma orizanol on inflammation and oxidative stress in skeletal muscle in obese rats

Abstract

Obesity is a disease that affects several organs, including skeletal muscle. Inflammation and oxidative stress are some of the mechanisms involved in muscle injury due to obesity and adipose tissue is the main organ that triggers these mechanisms. The objective of this work is to evaluate the preventive and treatment effect of ³-orizanol on inflammatory cytokines and oxidative stress products in the peripheral skeletal muscle of animals with obesity induced by a high carbohydrate diet. Male Wistar rats (n = 90), with 21 days of age, will be used, coming from the Vivarium of Universidade Estadual Paulista (UNESP). Four experimental groups (n = 15 animals / group) will be formed, according to figure 1 and will be studied in the period of 30 weeks. Groups G1, G2, G4 will not change diet during the experiment. The G1 group will receive standard diet, G2 standard diet + ³-orizanol, G4 high-carbohydrate diet (HCHO) + ³-orizanol. In the G1 and G2 groups, the effect of the diet on changes and obesity will be evaluated. In the G2 and G4 groups, the preventive effect of the compound will be evaluated. The animals in the G3 group will initially receive a high-carbohydrate diet and will undergo change and will receive the same diet plus ³-oryzanol after detecting the difference in body weight, approximately 20 weeks. Thus, we will be seeing the effect of treatment and also the carbohydrate-rich diet on the disease. At the end of the experiment, after the same fast of 12-15 hours, the animals will be anesthetized with sodium pentobarbital and euthanized by decapitation. Blood samples will be collected and plasma used for serum determinations of triacylglycerol, uric acid, adiponectin, insulin, TNF-± and interleukin-6 (IL-6) and the HOMA-IR index. In the soleus muscle and EDL will be measured IL-6, TNF-±, Malondialdehyde (MDA), Carbonylation, superoxide dismutase and catalase. Parametric data will be presented as mean ± standard deviation and comparisons between groups will be performed by analysis of parametric variance (ANOVA) and complemented with Tukey post-hoc test. Non-parametric data will be presented as median and interquartile (p25 and p75) and the test to be performed will be Kruskal-Wallis, complemented by Dunn's post-hoc test. Differences will be considered significant when p <0.05.
