Bispectral index, hemodinamic and respiratory effects of continuous infusion in differents rates of propofol in calves breathing spontaneously

Abstract

The aim of this study is to evaluate the bispectral index and the hemodynamic and respiratory effects of continuous rate infusion of propofol in calves. Eight Holtein calves with eight to 12 months of age, weighing 100-150 kg will be used. The animals will be induced to anesthesia with propofol at a dose of 3 mg/kg, intravenously and the calves trachea will be intubated with tracheal tubes of size compatible and positioned in right lateral recumbency. Immediately after the maintenance will be performed by continuous rate infusion of propofol, administered at two different rates, 0.6 mg kg-1 minute-1 and 0.8 mg kg-1 minute-1. In half of the animals (n=4) the experimental protocol will beginning with a rate of 0.6 mg kg-1 minute-1 during the first 40 minutes and after, the rate will be increased to 0.8 mg kg-1 minute-1 for more 40 minutes. In the other half of the animals (n=4) the experimental protocol will beginning with the highest rate (0.8 mg kg-1 minute-1) for 40 minutes and after it will be reduced (0.6 mg kg-1 minute-1) for 40 minutes, performing 80 minutes of propofol infusion in two different rates. Measurements of hemodynamic parameters (HR, SAP, DAP, MAP, CO, CVP, mPAP, PAPOm, CI, SI, SVR and PVR) and respiratory (ETCO2, f, SpO2, VT, VM, Raw, Cdyn and PPI), bloog gas analysis (PaO2, PaCO2, SaO2, DB, pH and HCO3) and bispectral index (BIS and EMG) will be evaluated before anesthesia (MB) and 20, 40, 60 and 80 minutes after continuous infusion rate of propofol (T20, T40, T60 and T80, respectively). Samples for measurement of propofol concentration in plasma will be carried out 20 minutes after initiation of continuous infusion of the drug (M20) and the remaining samples are collected at 20 minutes intervals for a period of 100 minutes (M40 a M120). Data from the BIS and EMG will be submitted to one-way ANOVA, and the others variables will be submitted to ANOVA followed by Bonferroni test (p<0.05). (AU)
