Evaluation of antimicrobial activity of essential oils on oral pathogens

Abstract

The oral cavity is inhabited by a mixed microbiota which, under determined circumstances, can overcome host defenses and act as opportunists producing some clinical manifestations. There is an interest in literature for natural products as sources of new bioactive molecules for the control of oral diseases, among these products there are essential oils and their secondary products due to their antimicrobial properties. The need to generate and add new information concerning the in vitro antimicrobial activity of essential oils, stimulated the development of this work, involving twenty species of the Germplasm Bank of the "Collection of Medicinal and Aromatic Plants - CPMA" UNICAMP. In this context, this research aims to identify the in vitro antimicrobial activity of essential oils and fractions of different species of medicinal and aromatic plants against oral pathogens, organized in planktonic and in biofilms forms, as well as identifying bioactive chemical classes present in essential oils and fractions of highest activity. Moreover, influence in the morphology of biofilms produced by essential oils and fractions selected will be evaluated which may help future studies on mechanisms of action. Also the toxicological potential will be verified in cell culture. Twenty fractions and essential oils extracted from plants belonging to the Germplasm Bank of the CPMA / CPQBA / UNICAMP will be evaluated for their antimicrobial activities against oral bacteria and yeast, using the following methods: determination of minimum inhibitory concentration (MIC) and bactericidal concentration and Fungicide Minimum (CBM / FCM) assessment of biofilm in initial phase and mature on bacterial viability (time kill), the occurrence of damage or changes in the morphology of microbial cells by scanning electron microscopy (SEM) and confocal scanning laser (CSLM); check antiproliferative activity on human skin fibroblasts ATCC CRL-2522TM, Vero cells, and tumor cell lines. Bioactive chemical classes present in the oils and fractions of higher activity and yield will be identified by RP-HPLC and GC-MS. This work will determine the antimicrobial potentiality of essential oils against oral microorganisms and provide additional information to the Germplasm Bank of CPQBA / UNICAMP and to the literature, to contribute to future studies regarding the mechanism of action, toxicology and chemical isolation of compounds may be promising for the generation of drug formulations for dental application.
