Melatonin and the control of energy metabolism: central and peripheral actions and the circadian timing of the metabolic function

Abstract

The aim of this work is to study the role played by the hormone melatonin in the control of energy metabolism. In particular, the role played by the central nervous system action of melatonin and the timing effect of melatonin signal regulating either the central master oscillator or the peripheral oscillators located in the metabolic active tissues. Using in vivo and in vitro experiments, in healthy or diabetic animals, in intact or pinealectomized, young or old animals, treated or not with melatonin, is our intention to study the metabolic function and gene expression in the central nervous system, white adipose tissue, skeletal muscle and the liver. On the other hand, is our aim to study the other side of the system, looking at the effects of the neuro-humoral signals linked to the several states of the metabolic function on the cellular and molecular processes involved in melatonin synthesis by the pineal gland. In addition, in collaboration with the Radiology Institute of Hospital of Clinics of the Faculty of Medicine, University of São Paulo, we intend to perform some clinical investigation trying to correlate the functional state of the pineal gland to alterations of the functional magnetic ressonance or positron emission tomography (PET) and SPECT images in healthy subjects and patients with diseases that might involve melatonin disfunction in their pathogenesis.
