Abstract

Snake venoms are the most studied animal toxins. The variability of venom composition was described not only intra- and interspecies but also according to sex, age and geographical distribution of the snakes, and in most studies the diet of snakes was considered as the triggering factor for this variation. The Herpetology Laboratory of the Butantan Institute provides venoms for antivenom production and for scientific research not only for the Institute, but to other institutions as well. Currently, it is observed that the number of snakes donated to the Institute has been steadily decreasing. This fact has stimulated the Herpetology Laboratory, where the snakes are kept, to develop technics to reproduce snakes in captivity and to become self-sufficient in creating these animals. Thus, the snake's colony of Herpetology Laboratory is composed mostly by animals born and/or kept in captivity for a long period of time and fed exclusively with mice and/or rats. Considering that the presence of changes in venom composition of snakes kept in captivity has not yet been investigated, the aim of this study is to evaluate the influence of diet on composition of B. jararaca and C. durissus terrificus snakes maintained in captivity in the Herpetology Laboratory of Butantan Institute. This evaluation involves the biochemical characterization of the venoms using classical methodology, such as protein measurement, polyacrylamide gel electrophoresis, including the identification of the proteins of interest by mass spectrometry, and high performance liquid chromatography. This study also includes the biological characterization of the samples, which comprise not only the determination of their well established activities, as proteolytic, coagulant and hemorrhagic, and the determination of lethal dose, but also "antivenomics" analysis and evaluation of the genotoxicity of toxins, activity which are attributed to the late manifestations of envenoming, through comet and micronucleus analyzes.
