Abstract

The chromatograph UPLC-MS / MS with accessories (including, among other items, detection system for tandem MS QTof, vacuum pump, LockSpray source with electrospray source, Lenovo digital monitor, ultra-performance chromatograph Acquity, arrangement detector diodes, MassLynx software, nitrogen generator, samples of chromatographic columns) should be used for the following purposes: a) analysis of the presence of antimicrobial peptides in coral samples Mussismilia spp. as well as microbial strains isolated from the same coral that these peptides should be uniquely identified by high resolution fragmentation analysis MS / MS; b) analyzing the presence of bryostatins and other complex polyketides present in Bugula spp., as well as microorganisms that possibly may be isolated and cultured from the bryozoan. These policetídios should also be clearly identified by high resolution fragmentation analysis MS / MS; c) analysis of the presence of tetrodotoxinas and derivatives in Tetraodon spp. (puffer fish) and micro-organisms associated to be isolated and identified from these fish. Toxins should also be identified by high resolution fragmentation analysis MS / MS; d) in dereplication experiments for unambiguous identification of known substances in micro-organisms and marine invertebrates extracts, seeking to identify those known so that we can prioritize the most isolating novel bioactive substances; e) identification of peptides Cnidaria, to be isolated by Prof. team André Tempone (Instituto Adolfo Lutz, SP). The main advantages of USE UPLC-UV-MS / MS are: ultra performance chromatographic separation, allowing the analysis with extreme accuracy the mass and UV spectrum of minority substances (often the most interested), use of different probes for ionization material crude (less clean) and equipment better prepared (as semi-purified samples) to investigate in detail the fragmentation tandem (MS / MS) in order to unequivocally identify the substances present in the samples, constructing a database for holding chemical profile analyzes, dereplication and identification of substances belonging to specific classes (bryostatins, tetrodotoxinas, polypeptides), exact mass measurements analysis to establish the atomic composition of unknown substances, possibility to use different ionization sources (ESI, APCI, CSE, APPI nanoFlow ESI) in a single apparatus for performing analyzes for different purposes aiming at the detection of the metabolic substances of different classes.
