Abstract

Mastitis is the most costly disease in dairy cows because it affects the production and milk quality, and also it reduces the industrial yield of dairy products. Coagulase-negative staphylococci (CNS) have become the most predominant pathogen causing bovine mastitis in many countries. The identification of most species of CNS is based on phenotypic characteristics, colony morphology and hemolytic potential, however, these conventional tests are expensive and time consuming. The objectives are to evaluate the effect of subclinical mastitis caused by CNS on milk yield and composition, and somatic cell count (SCC), as well as, the frequency of isolation of these microorganisms in mammary glands of dairy cattle. The aim is also to evaluate matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) for identification of CNS species. In total 30 infected quarters milk and 30 healthy contralateral quarters milk (control) will be evaluated by the selection from the samples collection of aseptic quarter milk of 120 cows for microbiological culture. Once isolated, the CNS species will be genotypically differentiated by the technique of polymerase chain reaction, associated with restriction fragment length polymorphism (PCR-RFLP) and by MALDI-MS. Once identified, the cows will be submitted to complete milking of individual mammary quarters, the production will be recorded and samples will be collected for analysis of CCS and composition.
