Abstract

The purpose of this project is developing a sandwich imunossensores array for multiplexed detection of pathogenic bacteria in foods using a microfluidic system. The pathogenic bacteria are the major causative agents of foodborne disease (FBD). The sensing device will be developed using primary antibodies immobilized on a film layer-by-layer (LBL) formed by gold nanoparticles (AuNPs) and políons on a graphite electrode array coupled to a microfluidic system. Massively modified magnetic particles with HRP, as electrochemical label, and the secondary antibodies are used to capture and pre-concentration of bacteria. As transduction system will evaluate the use of multiplex techniques voltammetry or chronoamperometry. For characterization of the films and the process of antibody immobilization techniques will be used for specular reflectance spectroscopy in the infrared, atomic force microscopy (AFM) and electrochemical impedance spectroscopy (EIS). The developed system will be evaluated using real samples spiked with DTA. The device will detect DTA at low detection limits, with high selectivity and low cost.
