Abstract This project aims to obtain peptides from bovine and buffalo whey proteins by enzymatic hydrolysis and apply them to in vitro gastrointestinal digestion simulation for dialyzability and culture of Caco-2 cells assays. The hydrolysates are obtained by using commercial proteases trypsin and pepsin immobilized on agarose, and corn cob powder and/or coconut fiber as alternatives low cost supports. Enzymatic hydrolysis with active and stabilized derivatives (enzyme linked to the support) under pH and temperature conditions predetermined will be conducted in fluidized bed reactor to make possible to easily separate the product from the reaction medium; the recycling and reuse of the derivatives will be also studied. For hydrolysis, the derivatives will be used in different combinations, allowing the release of different products. These will be later analyzed by chromatographic methods aiming the isolation of peptides. Once isolated, the peptides will undergo gastrointestinal conditions in order to verify its integrity. These studies could provide future projects to study possible biological and bioactive properties of these fragments and their applications in various food and pharmaceutical industries. | |
