Abstract

In this project we intend to develop and characterize a prophylactic vaccine strategy against dengue virus based on NS1 protein in combination with adjuvant. To achieve this, the recombinant NS1 protein DENV-2 and a truncated form of NS1 DENV 2 (DeltaC-NS1) will be expressed in E. coli and purified by metal affinity chromatography. Vaccine formulations will be prepared based on the NS1 and DeltaC-NS1 and administered as an antigen in BALB/c mice by intradermal route, in combination with the adjuvant poly (I:C). The cellular immune responses induced by anti-NS1 tested vaccine formulations will be monitored by testing as ICS (Intracellular Staining Citokine), ELISPOT (Enzyme-Linked Immunosorbent Spot) and measurement of cytotoxic activity in vivo. The humoral response induced anti-NS1 will be monitored by measurement of total IgG and IgG subclass anti-NS1 by ELISA. To evaluate the protective capacity conferred by the vaccine regimen, the mice will be challenged with a lethal dose of dengue virus. Moreover, the potential autoimmune responses induced by immunization with NS1 and DeltaC-NS1 will be measured by cross-reactivity with human serum albumin and human platelets in ELISA assays. Thus, at the end of the work, we expect to generate results and knowledge that will contribute to the development of an effective vaccine strategy against dengue virus. (AU)
