Abstract

This proposal aims to investigate the use of low temperature atmospheric pressure plasma (LTAPP) in Dentistry. The general aim is to develop protocols for the application of plasma and plasma activated liquid in vivo and ex vivo for diseases of dental interest and a portable equipment for clinical use. The specific aims are to develop effective protocols to the following relevant pathological conditions in Dentistry: oral candidosis, periodontitis, dental caries, endodontic infection and oral mucositis. Protocols of direct plasma application or plasma treated liquid (PTL, indirect application) will be tested. In case of infectious diseases, the effective parameters of LTAPP against each specific pathogenic biofilm will be standardized in vitro. After, these parameters will be evaluated for cyto and genotoxicity, using the most suitable cell line for each case. The effective (maximum antimicrobial effect) and safe (lowest toxicity) will be applied in vivo or ex vivo for validation. In relation to mucositis, the protocols of direct or indirect application will be evaluated in rats treated with 5-fluorouracyl, followed by histologic and immunohistochemical analyses. Systemic toxicity of PTL will be evaluated using Drosophila melanogaster model. The plasma device to be used in this study was developed and validated by the team of this project, with the support of several publications. For this reason, it is also aim of this project to transpose the current equipment to a small and portable prototype, maintaining its potentialities. (AU)
