Abstract

Streptococcus pneumoniae (pneumococcus) is a pathogenic, encapsulated bacteria. The capsules from these bacteria are constituted of a large chain polysaccharide (PS) and their different structures are related to a particular serotype. There are 90 different serotypes described in S. pneumoniae. Vaccines against pneumococcus are constituted by the purified PS, plain or conjugated to a protein. The advantage of a conjugated vaccine is to change the PS antigen from a thymus independent to thymus dependent one. The process for conjugate vaccine production is always very expensive particularly the conjugation step due to low recovery of conjugate. This is a special problem for pneumococcal vaccines which should include over 7 serotypes, which vary with geographical region. A variety of surface proteins have been investigated as alternative vaccine antigens, and pneumococcal surface protein A, PspA, has shown the most promising results. The present project intends to use PspA as carrier protein for PSs and antigen, aiming at extending the vaccine coverage using few conjugates. (AU)
