Antioxidants factors of bovine colostrum in cellular protection

Abstract

Among the components of bovine colostrum, the high presence of antioxidants factors is crucial to the health of the newborn, collaborating with overcoming oxidative stress after birth. In intensive fish farming, the stress due to common management practices results in lower reproductive performance and increased susceptibility to diseases. One of the main consequences of stress is the change of intestinal barrier permeability, increasing the absorption of unwanted substances from the lumen, and the supplementation of diet with antioxidants elements represents an important tool to minimize this condition. This study aims to determine the antioxidant potential of bovine colostrum and the influence of this milk secretion in the cellular protection and function of the intestinal barrier. The contribution of colostrum for overcoming stressful situations by fish species, using pacu as a model, and therefore the quality of the fish will also be evaluated. To determine the antioxidant potential of bovine colostrum, the chemical composition, concentration of antibodies (IgG), protein, enzymatic and non-enzymatic antioxidants and the oxygen radicals absorption ability will be analyzed in the colostrum of Holstein and Jersey primiparous and multiparous crossbred cows. Histophysiological responses will be evaluated in pacu (Piaractus mesopotamicus) under high stocking density (20 kg fish/m3) fed four diets containing 0, 10, 20 and 30% of bovine colostrum. Hematological and biochemical parameters, enzyme concentration of antioxidants and the oxygen radicals absorption ability will be analyzed in blood samples. The concentration of antioxidant enzymes, nucleotides and protein content, lipid peroxidation and the oxygen radicals absorption ability will be determined in the enteric, liver, muscle, kidney and gill tissue as well as liver glycogen. The cellular protection and function of the intestinal barrier will also be examined in the enteric tissue by optical and electron microscopy and western blotting. The fillet of pacu will be collected for the analysis of the quality of the fish fed with colostrum, being determined the chemical composition and physico-chemical stability of the sample. (AU)
