Prospection of chromosomal regions, genes and SNP related to Batida gait and genomic analysis of the population structure in horses of the brazilian breed Mangalarga Marchador

Abstract

The DMRT3 gene is described as the main gene involved in the determination of gait phenotypes in horses and the allele A of the 22999655C>A SNP has been reported as a causal variant of this trait. In the Mangalarga Marchador breed (MM), which exhibits two gait patterns with well-defined characteristics, genotypes AA and CA are associated with Picada gait and genotype CC with Batida gait. In this breed allele A of the DMRT3 gene is only related to the Picada gait. In a case-control Genome-wide association study (GWAS), recently conducted in FMVZ, Botucatu and FCAV, Jaboticabal-SP (Unesp) with the Equine SNP70 BeadChip (Illumina, Inc., USA) in Mangalarga Marchador, was demonstrated that contrary to what occurs with Picada gait, the Batida gait appears to be controlled by larger number of genes. However, due to the small number of animals (n = 48 n = 28 - Batida gait; n = 20 - Picada gait) and low chip density (65,157 SNP) used in this research, it was not possible to identify genomic regions significantly associated with the Batida gait. Based on the above, the initial objective of this research will be carried out, from larger sample of animals and higher density array, the prospection of genomic regions containing possible genes and SNP responsible for the phenotype of the Batida gait in the Mangalarga Marchador. To supplement will be analyzed the linkage disequilibrium (LD) and from this the effective population size and the occurrence of recent selection signatures in Mangalarga Marchador. To achieve the objectives, 192 horses (96 Batida gait and 96 Picada gait) recorded at studbook service of the Brazilian Association of Horse Breeders of Mangalarga Marchador (ABCCMM) will be genotyped with the Axiom Equine Genotyping Array (670,796 SNP: Affymetrix, USA). Thus, after performing imputation process between the genotypes of the animals analyzed with different chips, population studies and Bayesian GWAS (BayesB) will be conducted with 240 animals (124 of Batida gait and 116 of Picada gait). Blood samples of the animals will be collected during the National Exhibition of the breed, to be held in June 2017 in the city of Belo Horizonte-MG. The presence of full sibs will be avoided in the sample. (AU)
