Abstract

Trypanosomiases are diseases that affect humans, domestic and wild animals. In Brazil, especially in the last two decades, the livestock infection by Trypanosoma vivax has increasing incidence, affecting milk production. Clinical signs of the disease, such as anemia and weight loss, are generic and associated with aparasitemic intervals, makes the disease diagnosis challenging. Chemotherapeutic agents as diminazene diaceturate and isometamidium hydrochloride are available for disease treatment, but only the first is marketed in Brazil, although there are evidences of drug resistance. The isometamidium hydrochloride is being used improperly for the disease treatment in Brazil, which can lead to drug resistance. Although serological methods are useful, do not allow differentiating animals with active infection of successfully treated; therefore, molecular diagnostic methods are most suitable tools in outbreaks. The Loop-mediate Isothermal Amplification of DNA (LAMP) has been considered less influenced by the sample quality when compared with PCR, being a simple, quickly and more sensitive method, which does not require expensive laboratory resources. This study intends to evaluate the LAMP technique as a diagnosis tool and verify parasitological cure of naturally infected cattle with T. vivax after treatment with isometamidum, using 20 female cattle that will be treated with 1.0 mg/kg of isometamidium hydrochloride by intramuscular injection. Blood and serum samples will be collected on 0, 1, 2, 3, 4, 5, 6, 7, 15, 30, 60, 90 and 120 days after treatment, and then direct parasitological methods, ELISA, cPCR and LAMP will be performed, in order to detect T. vivax DNA and antibody response, knowledge of utmost importance for the implementation of properly containment measures in outbreaks.
